Subculturing: Protocol: Thawing of Hybridomas1. Add 7 ml of Dulbecco's modified Eagles medium (DMEM - e.g., Gibco-BRL Cat#1 1995-065) to a 15 ml tube.2. Quickly thaw one vial of cells in a 37C water bath.3. As soon as the ice melted, transfer the cell suspension into the tube with 7 ml DMEM and mix well.4. Spin the cells at 200 rpm in a clinical centrifuge for 3 mins.5. Aspirate the supematant.6. Resuspend the cells in 1 ml of DMEM with 10% fetal bovine serum (DMEN-10%FBS) and transfer it to ONE well of 6-well plate.7. Add an additional 2 ml of DMEM-10%FBS.8. Put the plate into a C02 incubator at 37C.9. 24 hrs later, examine the cells under microscope.10. If there are too many floating cells and/or dead cells, transfer the culture supernatant to another well of 6-well plate and add 3 ml of fresh DMEM-10% FBS to the original well.11. Incubate the cells in a C02 incubator at 37C.12. When the cells reach 75% confluence, split the cells 1:3 ? 1:5 in DMEM-10% FBS.Note:1. These hybridomas may attach to the dish pretty tight. For subculturing, you can mechanically wash up the cells by strong pipeting.2. These hybridomas do not like high cell density and viability drops significantly once over-confluence.
