ARP-173 is a human T-cell line, HUT 78, infected with Simian immunodeficiency virus (SIV-BK28). SIV-BK28 causes persistent infection and lymphadenopathy in rhesus macaques (Macaca mulatta). The plasmid clone pBK28-SIV is available as ARP-133.
Growth Characteristics: SIV-BK28 causes cellular atypia and giant cell formation occur in infected HUT 78 cells. The cells grow in single cell suspension with a doubling time of 3 days. Cultures should be passaged cells every 3 to 4 days. Seeding 5 × 106 cells in a 75cm2 flask yields approximately 18 × 106 to 20 × 106 cells within 1 week. The addition of fresh, uninfected cells is not necessary to maintain the culture. When thawing, immediately place vial in a 37°C water bath, transfer cells to a sterile 12 × 45 mm centrifuge tube. After 12 to 45 minutes, add fresh complete medium and centrifuge at 1200 RPM for 5 minutes. Please use the recommended growth medium, as the cells do not grow in other media.
Growth Medium: RPMI 1640 supplemented with 10% fetal bovine serum ARP-173 is negative for mycoplasma, bacteria and fungi. Each vial of ARP-173 contains approximately 9 x 106 cells in Minimal Essential Medium (MEM) supplemented with 50% fetal bovine serum and 10% dimethyl sulfoxide (DMSO).
