ARP-1446 is the CRFK (Crandell Reese Feline Kidney) cell line infected with feline immunodeficiency virus (FIV) AZR-1. Resistance was selected for entirely in vitro. Resistance is lost rapidly in the absence of AZT. The infected cells produce virus continually. The virus also replicates in feline lymphocytes and lymphoid cell lines.
Growth Characteristics: ARP-1446 is an epithelial-like, adherent cell line with a doubling time of 16 to 24 hours. Little syncytia formation is seen. Split when the culture becomes confluent. Medium should be changed at least twice weekly, but preferably every other day. High titers of virus can be obtained by collecting and replacing the culture medium from nearly confluent cells. These cells are very hardy and can be maintained in culture for long periods of time.
Growth Medium: A 1:1 mixture of Liebovitz (L-15) medium and DMEM supplemented with 10% fetal bovine serum, L-glutamine, 100 U per mL penicillin and 100 µg per mL streptomycin and 10 µM 3'azido-3'-deoxythymidine (azidothymidine; AZT).
ARP-1446 is negative for bacteria, Mycoplasma, yeast and mold.
Note: ARP-1446 should be frozen in culture medium containing AZT.
Each vial of ARP-1446 contains approximately 3.8 × 106 cells in a 1:1 mixture of Liebovitz (L-15) medium and DMEM supplemented with 10% fetal bovine serum and 10% dimethyl sulfoxide (DMSO) and 10 µM azidothymidine (AZT). Please refer to the appropriate data sheet for lot-specific information.
