ARP-401 is the H9 cell line infected with human immunodeficiency virus 1 (HIV-1) RF. The H9 cell line was derived from the human T-cell line, HUT 78, as a single clone. HIV-1 RF virus strain was obtained from peripheral blood lymphocytes in November 1983 and has been in continuous production in H9 cells since 1984. Strain RF is more cytopathic than HIV-1 IIIB.
Growth Characteristics: Cells grow as a single-cell suspension. Maintain cultures at 5 × 105 to 1 × 106 cells per mL for optimum growth and split every 2 to 3 days at a ratio of 1:2 to 1:4. When cells are first started in culture, split at 1:2 only for the first week. The HIV-1 p24 antigen capture assay shows a p24 level of approximately 5 ng per mL. Since the amount of p24 is not very high, adding uninfected H9 cells (ARP-87) to the culture can increase the virus output.
Growth Medium: RPMI 1640 supplemented with 10% fetal bovine serum and 2 mM L-glutamine ARP-401 is negative for Mycoplasma, bacteria and fungi. Each vial of ARP-401 contains approximately 1 x 107 cells in RPMI 1640 supplemented 40% fetal bovine serum and 10% dimethyl sulfoxide (DMSO).
