ARP-2955 is an indicator cell line infected with the amphotropic retroviral vector LT4SN to express human CD4, transfected with a plasmid containing the human immunodeficiency virus 1 (HIV-1) long terminal repeat (LTR) fused to the ß-galactosidase gene. ARP-2955 allows for the rapid quantification of multiple strains of infectious Simian immunodeficiency virus (SIV).The parental rhesus macaque mammary tumor line CMMT (ATCC ® CRL-6299™) expresses Simian type D retrovirus (SRV-1); both CMMT and sMAGI cells have high endogenous reverse transcriptase (RT) activity due to SRV-1 expression.
CMMT-CD4 cells were generated by infecting the parental CMMT cell line with the amphotropic retroviral murine leukemia virus (MuLV)-based vector PA317. Cells expressing high levels of CD4 were selected by flow cytometry and fluorescence-activated cell sorting (FACS).
CMMT-CD4-LTR-ß-Gal (sMAGI) indicator cells were constructed by transfecting CMMT-CD4 cells with the plasmid pEQ447 containing the hygromycin resistance gene under control of the CMV major-immediate promoter, the ß-Gal gene downstream of a truncated HIV-1 LTR, and the Simian vacuolating virus 40 (SV-40) T antigen nuclear localization signal.
