ARP-4692 is derived from the lentiviral packaging vector pHP-1dl.28 (refer to attached figure from Gene Ther.) with an additional 695 base pair env deletion (NdeI-AseI). This lentiviral helper construct generates HIV-1 Gag-Pol, Tat, Rev, Vpr, Vif, and Vpu. It is derived from pUC18-based pNL4-3 with deletions of 5', U3, 5'UTR, part of env, nef and the 3' LTR. This construct is 11,729 base pairs including the insert and is ampicillin resistant. Bacteria transformed with this construct are prone to deletions when grown at 37°C with vigorous shaking. Cultures should be grown at 32°C with gentle overnight shaking. This construct can be propagated in DH5α, Top-10, HB101 or STBL2. Each vial of ARP-4692 contains approximately 5 µg of dried purified DNA stabilized in DNAstable®PLUS. Please refer to the appropriate data sheet for lot-specific information.
