  Question How should host cells be prepared for human metapneumovirus (hMPV) inoculation?

  Answer The hMPV isolates in BEI Resources are grown in LLC-MK2 Derivative cells (ATCC® CCL-7.1™). The cells should be adapted to low serum conditions by growing them in Opti-MEM Minimal Essential Medium supplemented with 2% fetal bovine serum and 2mM L-glutamine. It is also important to use low-passage number cells as they will need to be adapted to this low serum condition prior to inoculation. When the cells are ready to be used for inoculation, the monolayer should be rinsed with serum free medium or Dulbecco’s phosphate buffered saline prior to inoculation to remove any residual fetal bovine serum.
